nab-sirolimus is a nanoparticle albumin-bound formulation of sirolimus (also known as rapamycin), a macrocyclic lactone produced by Streptomyces hygroscopicus. It acts as a potent inhibitor of the mammalian target of rapamycin (mTOR), which regulates cell growth and proliferation. By inhibiting mTOR, nab-sirolimus exerts antitumor activity and can potentiate the cytotoxicity of other chemotherapeutic agents. The albumin-binding allows for enhanced delivery to tumor cells via albumin-mediated endocytosis[5][8]. lomustine is an oral alkylating agent in the nitrosourea class, used primarily for brain tumors and refractory or relapsed Hodgkin's lymphoma. It works by alkylating DNA and RNA, causing cross-linking at the O6 position of guanine bases, leading to inhibition of DNA synthesis and cell death[1][3][6]. The combination is under investigation for synergistic effects in cancer therapy.
