nab-Sirolimus (ABI-009, also known as nab-rapamycin) is a human albumin-bound nanoparticle formulation of sirolimus, a potent inhibitor of the mechanistic target of rapamycin (mTOR). Temozolomide is an oral alkylating agent that methylates DNA and induces tumor cell death. The combination has been studied in pediatric patients with relapsed or refractory solid or central nervous system tumors. Nab-sirolimus acts by inhibiting mTOR signaling, thereby blocking cell growth and proliferation, while temozolomide exerts cytotoxic effects through DNA methylation. The combination aims to enhance antitumor efficacy by targeting both cellular proliferation pathways and directly damaging tumor DNA[2][3][5].
