Overview

Nab-thiocolchicine dimer (ABI-011) is a nanoparticle albumin-bound formulation of a thiocolchicine dimer, developed using proprietary nab technology. It is a dual-action antineoplastic agent that functions as both a microtubule inhibitor (antitubulin) and a DNA topoisomerase I (TOP1) inhibitor. In addition to its cytotoxic effects on dividing cancer cells, it acts as a vascular disrupting agent (VDA), targeting and collapsing established tumor vasculature to induce rapid secondary tumor cell death. Originally developed by Abraxis BioScience, the drug was designed to enhance the delivery and therapeutic profile of the thiocolchicine dimer. Preclinical studies demonstrated significant anti-tumor activity in various mouse xenograft models, including breast, colon, prostate, and ovarian carcinomas.
