Nabi Anti-D is a sterile, freeze-dried gamma globulin (IgG) fraction derived from human plasma, containing high titers of antibodies to the Rho(D) antigen (D antigen) of human red blood cells. Developed by Nabi Biopharmaceuticals, it was primarily indicated for the suppression of Rh isoimmunization in Rho(D)-negative individuals exposed to Rho(D)-positive blood, such as during pregnancy or delivery of an Rh-positive infant, to prevent hemolytic disease of the newborn in subsequent pregnancies. It was also indicated for the treatment of non-splenectomized Rho(D)-positive patients with chronic or acute Immune Thrombocytopenic Purpura (ITP). In Rh prophylaxis, the drug works by coating fetal Rh-positive red blood cells in the maternal circulation, facilitating their clearance before the maternal immune system can mount an antibody response. In the context of ITP, it acts by competitively inhibiting Fc gamma receptors on splenic macrophages, thereby reducing the destruction of antibody-coated platelets. The product was part of the biologics portfolio sold by Nabi to Biotest Pharmaceuticals in 2007.
