Overview

Nadecnemab is a human IgG4κ monoclonal antibody developed by Regeneron Pharmaceuticals. It functions as an antagonist of glial cell line-derived neurotrophic factor family receptor alpha-3 (GFRα3). The drug was investigated for its potential in treating chronic pain conditions, particularly osteoarthritis (OA) knee pain, by targeting the artemin-GFRα3 signaling pathway, which has been implicated in various painful conditions. While Nadecnemab demonstrated safety comparable to placebo in a Phase 1 study in healthy volunteers, a subsequent Phase 2 study in patients with OA knee pain failed to meet its primary and secondary efficacy endpoints.
