Overview

Nadifloxacin is a topical fluoroquinolone antibiotic primarily used for the treatment of acne vulgaris and various bacterial skin infections. It exhibits broad-spectrum antibacterial activity against aerobic Gram-positive, Gram-negative, and anaerobic bacteria—including methicillin-resistant Staphylococcus aureus (MRSA) and Cutibacterium acnes. Its mechanism of action involves inhibition of bacterial DNA gyrase and topoisomerase IV, enzymes essential for DNA replication and transcription in bacteria. This dual inhibition disrupts bacterial DNA synthesis, leading to bactericidal effects[1][3][8]. Nadifloxacin also demonstrates anti-inflammatory properties by inhibiting proinflammatory cytokine production in immune cells and keratinocytes[6]. The drug is approved for use in Japan and several other countries as a topical agent for acne vulgaris and skin infections.
