Nafabelltan (CKD-314) is an intravenous formulation of nafamostat mesylate, a synthetic serine protease inhibitor, being developed by Chong Kun Dang Pharmaceutical for the treatment of COVID-19. Originally approved in Japan and South Korea for pancreatitis and disseminated intravascular coagulation, nafamostat was identified as a potent inhibitor of the host cell surface protease Transmembrane Protease Serine 2 (TMPRSS2). By blocking TMPRSS2, Nafabelltan prevents the SARS-CoV-2 spike protein from being cleaved and activated, thereby inhibiting viral entry into human lung cells. The drug has undergone Phase 2 testing in Russia and Phase 3 trials in South Korea, specifically targeting hospitalized patients with severe COVID-19 pneumonia.
