Nafazatrom is a synthetic small molecule pyrazolinone derivative developed as a novel antithrombotic and anti-metastatic agent. It functions by stimulating prostacyclin (PGI2) production and inhibiting lipoxygenase enzymes, which are involved in arachidonic acid metabolism. Nafazatrom has been evaluated for cardioprotective and anti-metastatic activity, especially in advanced cancer and coronary artery disease. Clinical studies found nafazatrom to be safe and well-tolerated but largely lacking in efficacy for cancer and cardiovascular endpoints. Mechanistically, nafazatrom modulates prostaglandin production and may influence platelet function and monocyte arachidonate metabolism, although these effects did not consistently correlate with antitumor response in clinical settings[1][3][4][6][7][10].
