Overview

Naftazone is a naphthoquinone derivative small molecule with dual therapeutic applications. Historically, it has been utilized as a vasoprotective and venotonic agent for the treatment of vascular conditions such as chronic venous insufficiency, varicose veins, and hemorrhoids. In this context, it functions by inhibiting phosphodiesterase enzymes, which increases cyclic AMP levels in vascular smooth muscle cells to improve venous tone and capillary resistance. It also exhibits anti-inflammatory properties by inhibiting mediators like histamine and serotonin. More recently, naftazone has been repositioned under the code name CVXL-0107 for the treatment of Parkinson's disease and levodopa-induced dyskinesia. Its mechanism in the central nervous system involves the inhibition of glutamate release, which is intended to mitigate excitotoxicity and motor complications associated with advanced Parkinson's disease.
