Overview

Naftifine organogel is a topical antifungal formulation developed by MediQuest Therapeutics for the treatment of onychomycosis, specifically distal subungual tinea unguium. Naftifine is an allylamine antifungal agent that acts by inhibiting the enzyme squalene epoxidase (also known as squalene 2,3-epoxidase). This inhibition blocks the conversion of squalene to squalene epoxide, a critical step in fungal ergosterol biosynthesis. The resulting depletion of ergosterol and the toxic accumulation of intracellular squalene disrupt the fungal cell membrane, leading to a fungicidal effect against dermatophytes. The MediQuest formulation utilizes a novel organogel vehicle designed to facilitate the penetration of naftifine through the nail plate to reach the infected nail bed. The development program included Phase 2b clinical trials evaluating 2% (MQX-5858) and 6% (MQX-5866) concentrations.
