NAI003-7 is a first-in-class small-molecule endoplasmic reticulum (ER) stress modulator developed by Nain Biotech. It is designed to dually target Protein Disulfide Isomerase Family A Member 6 (PDIA6), Inositol-Requiring Enzyme 1 (IRE1), and Histone Deacetylases (HDACs). The compound selectively induces regulated cell death in cancer cells harboring STK11 and TP53 genetic and functional defects, which are common in hard-to-treat malignancies such as pancreatic cancer. By modulating ER stress pathways and inhibiting HDAC activity, NAI003-7 exploits metabolic and redox vulnerabilities in these specific tumor types while maintaining a favorable safety profile in normal human cells.
