Overview

Naive dendritic cells (naive DCs) are a subtype of dendritic cells, which are professional antigen-presenting cells (APCs) central to the initiation of immune responses. These cells are termed "naive" because they have not yet encountered or processed antigen for presentation to T cells. In their immature or naive state, dendritic cells are highly phagocytic and efficient at sampling their environment, but they express low levels of costimulatory molecules and MHC complexes. Upon encountering antigens and receiving activation signals, naive DCs undergo a maturation process, upregulating costimulatory molecules and MHC, migrating to lymphoid tissues, and becoming highly potent in priming naive T cells (both CD4+ and CD8+) to initiate adaptive immune responses[3][7]. This property underlies their key role as a modality in immunotherapy, where naive or minimally manipulated DCs are loaded with antigens ex vivo and reinfused as cellular vaccines to induce anti-tumor immunity[1][5]. Major current therapeutic investigation areas include cancer immunotherapy and vaccine development.
