The naked DNA E7 vaccine is a therapeutic DNA-based immunotherapy designed to treat malignancies and pre-cancerous lesions caused by Human Papillomavirus (HPV) type 16, most notably cervical cancer. It consists of a recombinant bacterial plasmid that encodes the HPV-16 E7 oncoprotein, a primary driver of viral oncogenesis. The term "naked" refers to the delivery of the DNA plasmid in a simple aqueous buffer (such as phosphate-buffered saline) without the use of viral vectors, lipid nanoparticles, or complex chemical carriers. Upon administration, typically via intramuscular or intradermal injection, the plasmid is internalized by host cells, which then express the E7 protein. This endogenous production of the viral antigen allows for processing and presentation via MHC class I molecules, stimulating a robust cellular immune response. Specifically, it aims to activate CD8+ cytotoxic T lymphocytes (CTLs) to recognize and eliminate E7-expressing tumor cells. While naked DNA vaccines are known for their safety and ease of manufacture, they often exhibit limited immunogenicity in humans, leading researchers to explore physical delivery enhancements like electroporation or chemical delivery systems like chitosan nanoparticles.
