Drug intelligence / Profile preview
NaKtide is a 20-amino-acid mimetic peptide derived from the nucleotide-binding (N) domain of the Na/K-ATPase α1 subunit. It acts as a potent antagonist of Na/K-ATPase signaling by binding to the kinase domain of Src family kinases (specifically c-Src), thereby preventing Src activation and disrupting the Na/K-ATPase/Src receptor complex. This inhibition blocks the feed-forward Na/K-ATPase oxidant amplification loop (NKAL), reducing systemic oxidative stress, inflammation, and lipid accumulation. NaKtide and its cell-permeable derivative, pNaKtide (conjugated to an HIV-Tat leader sequence), have been investigated in preclinical models for the treatment of obesity, metabolic syndrome, atherosclerosis, uremic cardiomyopathy, chronic kidney disease-associated anemia, and neovascularization disorders of the eye.
