Nal-anti TIGIT is a bifunctional antibody-drug conjugate (ADC) developed by TuHURA Biosciences for the treatment of myelodysplastic syndromes (MDS). This prototype ADC combines a small molecule Delta receptor inhibitor with an anti-TIGIT antibody. The drug is designed to target myeloid-derived suppressor cells (MDSCs), which are known to suppress T and NK cell function and contribute to immune evasion in the tumor microenvironment. By inhibiting the Delta receptor on MDSCs and blocking the TIGIT checkpoint pathway, nal-anti TIGIT aims to modulate immunosuppression, overcome resistance to checkpoint inhibitors, and enhance antitumor immunity. Its primary indication is high-risk myelodysplastic syndromes[1][2].
