Overview

Nalbuphine + magnesium sulfate is a combination of two agents commonly used as adjuvants in anesthesia and pain management, particularly in perioperative and obstetric settings. Nalbuphine is a synthetic opioid analgesic with mixed agonist-antagonist activity—acting as an agonist at kappa opioid receptors and an antagonist at mu opioid receptors—providing potent analgesia with a ceiling effect on respiratory depression. Magnesium sulfate, an inorganic salt, acts primarily as an NMDA receptor antagonist and has been shown to enhance the efficacy of other analgesics while providing additional benefits such as reducing shivering during spinal anesthesia. The combination is studied for its ability to improve postoperative pain control, prolong the duration of anesthesia, reduce perioperative shivering, and potentially decrease the need for additional opioids[1][6][8].
