Overview

Nalcol is a sustained-release oral formulation of naloxone developed by S.L.A. Pharma AG for the treatment of opioid-induced constipation (OIC) in patients with persistent non-cancer pain. As a mu-opioid receptor antagonist, naloxone works by blocking opioid receptors in the gastrointestinal tract. The slow-release formulation is designed to provide local action in the gut while maintaining low systemic bioavailability due to significant hepatic first-pass metabolism. This allows the drug to alleviate constipation without compromising the central analgesic effects of the opioids being taken for pain management.
