Naldebain + SafeTynadol is a combination analgesic therapy being developed by Sinew Pharma Inc. for the management of moderate to severe postoperative pain, specifically following knee replacement surgery. The combination consists of Naldebain (sebacoyl dinalbuphine ester), a long-acting prodrug of the opioid analgesic nalbuphine, and SafeTynadol (SNP-810), a liver-safe formulation of acetaminophen. Nalbuphine acts as a kappa-opioid receptor agonist and a mu-opioid receptor antagonist, providing potent analgesia with a lower risk of respiratory depression and addiction compared to traditional mu-agonists. SafeTynadol is designed to provide the analgesic and antipyretic benefits of acetaminophen while utilizing specific additives (mannitol and sucralose) to inhibit the metabolic pathways that lead to hepatotoxic metabolites like NAPQI. This combination aims to provide synergistic pain relief with an improved safety profile.
