NALIRIFOX + adebrelimab

NALIRIFOX + adebrelimab is an investigational chemo-immunotherapy combination regimen being evaluated for the first-line treatment of locally advanced unresectable or metastatic pancreatic ductal adenocarcinoma (PDAC). NALIRIFOX is a quadruplet chemotherapy regimen consisting of liposomal irinotecan (a topoisomerase I inhibitor), oxaliplatin (a platinum-based DNA crosslinking agent), 5-fluorouracil (a thymidylate synthase inhibitor), and leucovorin (a biochemical modulator of 5-FU). Adebrelimab (SHR-1316) is a humanized monoclonal antibody that targets the programmed death-ligand 1 (PD-L1) immune checkpoint, designed to restore anti-tumor T-cell activity. In clinical trials, this combination is often paired with Personalized Ultra-fractionated Stereotactic Adaptive Radiotherapy (PULSAR) to potentially enhance the systemic immune response through local tumor control and the abscopal effect.
