Overview

Nalotimagene carzoleucel (Zalmoxis) is a cell-based gene therapy consisting of allogeneic T cells genetically modified with a retroviral vector encoding a truncated form of the human low-affinity nerve growth factor receptor (ΔLNGFR) and the herpes simplex virus type 1 thymidine kinase (HSV-TK Mut2) suicide gene. Developed by MolMed, it is indicated as an adjunctive treatment in adult patients with high-risk hematological malignancies undergoing haploidentical hematopoietic stem cell transplantation (HSCT). The suicide gene system allows for the selective elimination of the infused lymphocytes using ganciclovir or valganciclovir in the event of severe graft-versus-host disease (GvHD), thereby supporting immune reconstitution while providing a safety switch to control GvHD.
