Overview

Namodenoson is an orally available small molecule drug, also known as CF102 and Cl-IB-MECA. It acts as a highly specific and selective agonist of the A3 adenosine receptor (A3AR). Namodenoson exhibits anti-cancer and anti-inflammatory properties, with its primary mechanism involving the deregulation of NF-κB and Wnt signaling pathways. This leads to apoptosis in tumor cells and inhibition of inflammatory cytokines (such as TNF-α, IL-1, IL-6, IL-8), while stimulating positive cytokines like G-CSF and adiponectin. These effects result in reduced liver inflammation, steatosis, fibrosis (notably in metabolic dysfunction-associated steatohepatitis/MASH), and tumor growth inhibition (notably in hepatocellular carcinoma/HCC). Namodenoson is being developed by Can-Fite BioPharma for indications including advanced hepatocellular carcinoma (HCC), pancreatic cancer, and metabolic dysfunction-associated steatohepatitis (MASH)[2][5][6][7][8].
