NAMPT siRNA is a small interfering RNA (siRNA) designed to target and silence the expression of the Nicotinamide phosphoribosyltransferase (NAMPT) gene. NAMPT is the rate-limiting enzyme in the NAD+ salvage pathway, which is often upregulated in the tumor microenvironment (TME) of multiple myeloma (MM). By knocking down NAMPT expression, particularly in tumor-associated macrophages, this agent depletes intracellular NAD+ pools, leading to mitochondrial dysfunction and the inhibition of the SIRT1-STAT3-SPP1 signaling axis. This molecular disruption shifts macrophages toward a pro-inflammatory phenotype and reduces the expression of immunosuppressive factors like SPP1, thereby enhancing CD8+ T-cell cytotoxicity and resensitizing myeloma cells to proteasome inhibitors like bortezomib.
