Nano-273 is an albumin-based nanoformulation of a dual-targeting small molecule designed to simultaneously activate the Stimulator of Interferon Genes (STING) pathway and inhibit Phosphoinositide 3-kinase gamma (PI3Kγ). This dual mechanism is intended to overcome the complex immune suppression found in pancreatic cancer, where both myeloid cells and regulatory B cells (Bregs) contribute to a pro-tumor environment. By activating STING, Nano-273 stimulates myeloid cells to promote anti-tumor immunity; by inhibiting PI3Kγ, it prevents the STING-induced expansion of Bregs that would otherwise counteract the immune activation. Preclinical studies in transgenic KPC mice have demonstrated that Nano-273 effectively delivers its payload to tumors and lymph nodes, and when used in combination with anti-PD-1 therapy, it can achieve durable efficacy.
