Drug intelligence / Profile preview
Nano-combretastatin is a dendrimer-based nanoparticle conjugate of the vascular disrupting agent (VDA) combretastatin A-4 (CA4). Developed by researchers at Henry Ford Hospital, this formulation was engineered to overcome the clinical limitations of CA4 and its prodrug combretastatin phosphate (CA4-P), which include poor systemic bioavailability, non-specific distribution, and a short blood half-life. As a VDA, nano-combretastatin targets the established and newly formed vasculature of solid tumors by binding to the colchicine site of tubulin, leading to microtubule depolymerization and rapid vascular collapse. Preclinical studies in rat glioma models have demonstrated that intravenous administration of nano-combretastatin induces significant central necrosis within the tumor core, which can be effectively monitored using MRI-determined tissue parameters such as blood flow and distribution volume.
