Nano-DC vaccine is an experimental autologous dendritic cell-based cancer vaccine developed by the Houston Methodist Research Institute for the treatment of HER2-positive breast cancer. The vaccine consists of bone marrow-derived dendritic cells (DCs) loaded with porous silicon microparticles (PSMs) that are packaged with HER2 antigen peptides. The PSMs serve a dual role: they act as a reservoir for the sustained release of HER2 antigen peptides to enhance cross-presentation, and they function as a potent adjuvant by stimulating type I interferon (IFN) expression in DCs via the TRIF/MAVS pathways. In preclinical murine models of HER2-positive breast cancer, the Nano-DC vaccine has been shown to activate and expand antigen-specific CD8+ cytotoxic T cells and promote a Th2-to-Th1 transition in the tumor microenvironment, leading to robust anti-tumor activity.
