NANO-Lopinavir

NANO-Lopinavir is an experimental solid drug nanoparticle (SDN) formulation of the HIV-1 protease inhibitor lopinavir. Developed through a collaboration between the University of Liverpool, St Stephens AIDS Trust, and Chelsea & Westminster Hospital, this formulation utilizes nanotechnology to enhance the oral bioavailability of lopinavir. By improving drug delivery, NANO-Lopinavir aims to achieve therapeutic drug exposure at significantly lower doses—potentially a 50% reduction—compared to conventional formulations like Kaletra (lopinavir/ritonavir). It has been evaluated in Phase 1 pharmacokinetic studies in healthy volunteers, where it was administered as powder-filled capsules, often in combination with ritonavir as a pharmacokinetic booster to maintain therapeutic levels.
