nano-miR-34a is an experimental cancer therapeutic consisting of a miR-34a mimic encapsulated within a self-assembled nanovector delivery system. Developed by researchers at the University of Kansas, this drug is designed to restore the levels of the tumor suppressor microRNA miR-34a, which is frequently lost or downregulated in cancers with p53 mutations. miR-34a functions by targeting and downregulating key oncogenic pathways, specifically the Bcl-2 and Notch signaling pathways, which are critical for the survival, self-renewal, and chemoresistance of cancer stem cells (CSCs). Preclinical studies in pancreatic, breast, and prostate cancer models have demonstrated that systemic administration of nano-miR-34a can effectively deliver the miRNA to tumors, reduce CSC populations, and inhibit tumor growth and tumorsphere formation.
