Overview

nano-TriNKE is a nanoparticle-based immunotherapy platform designed to target EGFR-overexpressing tumors while recruiting and activating natural killer (NK) cells. The platform is constructed using biocompatible PEG-PLGA nanoparticles functionalized with three distinct antibodies: cetuximab (targeting EGFR), anti-CD16, and anti-4-1BB. This trispecific configuration enables the simultaneous engagement of tumor cells and the activation of NK cells through both the CD16 and 4-1BB costimulatory pathways. Furthermore, the nano-TriNKE platform can encapsulate cytotoxic agents such as epirubicin, allowing for a combined chemo-immunotherapeutic approach. Developed as a research-stage biologic, it was first detailed in a 2020 publication in Science Advances.
