NanoDOPA is a self-assembled nanodrug developed as a prodrug formulation of levodopa (l-DOPA) for the treatment of Parkinson's disease. It is synthesized from an amphiphilic block copolymer featuring poly(l-DOPA(OAc)2) as the hydrophobic segment. This nanostructure allows for the gradual enzymatic hydrolysis and liberation of l-DOPA under physiological conditions, aiming to maintain stable plasma levels and avoid the pulsatile stimulation of dopamine receptors that leads to levodopa-induced dyskinesia (LID). Preclinical studies in MPTP-induced Parkinson's disease mouse models have shown that NanoDOPA significantly improves motor symptoms without the side effect of dyskinesia and exhibits reduced systemic toxicity compared to conventional l-DOPA.
