Overview

nanoparticle-siRNA (CD133) refers to a therapeutic platform designed to target cancer stem cells (CSCs) by delivering small interfering RNA (siRNA) against the CD133 (Prominin-1) antigen using nanoparticle carriers. CD133 is a transmembrane glycoprotein frequently used as a marker for CSCs in various solid tumors, including glioblastoma, colorectal cancer, and hepatocellular carcinoma. These CSCs are often resistant to standard chemotherapy and radiotherapy, contributing to tumor recurrence and metastasis. The nanoparticle delivery system—which may consist of lipids, polymers, or inorganic materials—protects the siRNA from degradation in the bloodstream and facilitates its uptake into target cells. Once inside, the siRNA triggers the RNA interference (RNAi) pathway to silence CD133 expression, thereby reducing the self-renewal capacity and tumorigenicity of the CSC population. This approach is currently in the preclinical stage of development, with numerous studies demonstrating its potential to sensitize tumors to conventional treatments.
