Overview

**Nanoquinacrine** is a nanoparticle formulation of quinacrine developed to enhance its anti-cancer effects, especially against cervical cancer cells and cancer stem cells. Compared to conventional quinacrine, nanoquinacrine demonstrates increased cytotoxicity and efficacy in preclinical models. Its mechanism of action involves inhibiting the Hedgehog-GLI (HH-GLI) signaling pathway by directly binding to the GLI1 DNA complex, leading to downregulation of tumor-promoting GLI targets, increased expression of negative regulators such as GSK3β and PTEN, and induction of apoptosis in cancer stem cells. This nanoformulation addresses pharmacokinetic limitations of quinacrine, improving drug delivery and tumor targeting[1][4].
