Nanosomal docetaxel lipid suspension (NDLS) is a novel lipid-based nanoformulation of the semi-synthetic taxane chemotherapeutic agent docetaxel. Developed to overcome toxicity issues associated with conventional formulations that use polysorbate 80 and ethanol as excipients, NDLS eliminates these components, thereby reducing the risk of hypersensitivity reactions and infusion-related toxicities. This allows administration without corticosteroid premedication. The nanosomal formulation (<100 nm particles) enhances water solubility and may improve drug delivery to tumors via enhanced permeability and retention effects in compromised tumor vasculature. NDLS is approved in India for several cancers including breast cancer, hormone-refractory prostate cancer (such as metastatic castration-resistant prostate cancer), locally advanced squamous cell carcinoma of the head and neck, non-small cell lung cancer, and advanced gastric adenocarcinoma[2][3][4][5]. Docetaxel acts by binding to microtubules in cells, stabilizing them against depolymerization which disrupts mitosis and leads to apoptosis[5].
