**NanoSTING-SN** is a preclinical intranasal, multi-antigen protein-subunit vaccine designed to prevent coronavirus infection, particularly COVID-19. It combines trimeric SARS-CoV-2 spike protein and SARS-CoV-2 nucleocapsid protein with NanoSTING, a liposomal nanoparticle adjuvant containing the endogenous STING agonist 2'-3' cGAMP. Local activation of STING in the nasal respiratory tract promotes innate antiviral signaling, while the spike and nucleocapsid antigens induce mucosal and systemic adaptive immunity. In animal studies, NanoSTING-SN eliminated detectable SARS-CoV-2 replication in lungs and nostrils after Delta challenge and prevented transmission of Omicron BA.5 to naïve hamsters; it also generated cross-reactive immune responses against related coronaviruses.
