The NANT Neoadjuvant TNBC Vaccine is an investigational, molecularly informed multi-agent neoadjuvant immunotherapy regimen developed by ImmunityBio for previously untreated stage II or III triple-negative breast cancer. In the withdrawn Phase 2 QUILT-3.057 study, the regimen combined metronomic cytotoxic chemotherapy, antigen-directed yeast and adenoviral-vector therapeutic cancer vaccines, the IL-15 superagonist ALT-803, haNK allogeneic natural killer cells, and the anti-PD-L1 antibody avelumab. Its intended strategy was to promote immunogenic tumor-cell death, induce antigen-specific T-cell responses, activate NK and T cells, and relieve PD-L1-mediated immune suppression before surgery.
