The NANT Pancreatic Cancer Vaccine is a multi-modal, combination immunotherapy regimen in development for the treatment of pancreatic cancer, especially for patients who have progressed on or after standard-of-care therapies. The approach integrates low-dose (metronomic) chemoradiation to induce damage-associated molecular pattern (DAMP) signals and expose tumor antigens, delivery of tumor antigen vaccines (adenoviral, yeast-based, and fusion-protein vaccines targeting KRAS and other tumor antigens), stimulation of natural killer (NK) and T-cells using the IL-15 superagonist N-803 (Anktiva), and administration of engineered, PD-L1-targeted, high-affinity off-the-shelf NK cell therapy (PDL1-t-haNK cells). The therapy aims to coordinate engagement of both the innate and adaptive immune systems to induce durable anti-tumor responses. The regimen is being tested in clinical trials (notably the QUILT 88 and QUILT 3.070 studies) in metastatic pancreatic cancer and is developed by ImmunityBio, building on earlier work by NantKwest.
