**NANT Squamous Cell Carcinoma Vaccine** is an investigational, molecularly informed multi-agent cancer-immunotherapy regimen developed for advanced squamous cell carcinomas progressing after platinum-based chemotherapy and PD-1 or PD-L1 blockade. The QUILT-3.090 protocol combined metronomic chemotherapy and tumor-directed treatment with adenoviral and recombinant yeast cancer vaccines, the IL-15 superagonist nogapendekin alfa inbakicept, and allogeneic high-affinity natural killer cells. Vaccine elements were designed to elicit antigen-specific T-cell responses against carcinoembryonic antigen, HER2, brachyury, MUC1, and mutant RAS, while the broader regimen aimed to augment innate NK-cell and adaptive antitumor immunity. It was investigated in squamous cell carcinoma, including head and neck squamous cell carcinoma and squamous non-small cell lung cancer, but is not an approved product.
