NANTIL-15 (New ANTagonist of IL-15) is a recombinant fusion protein designed to selectively inhibit the pro-inflammatory actions of interleukin-15 (IL-15). It consists of a human IL-15 mutein containing three specific mutations (L45D, N65K, and L69R) fused to an Fc immunoglobulin domain. The N65K and L69R mutations are strategically placed to abrogate the recruitment of the IL-2Rβ (CD122) receptor chain, while the molecule retains high-affinity binding to the IL-15Rα (CD215) chain. This selectivity allows NANTIL-15 to block IL-15 signaling through the high-affinity trimeric receptor complex (IL-15Rα/IL-2Rβ/γc) without interfering with IL-15 or IL-2 signaling through the dimeric IL-2Rβ/γc receptor, which is essential for the homeostasis of natural killer (NK) and memory CD8+ T cells. Developed by researchers at Inserm and CNRS, NANTIL-15 has demonstrated efficacy in reducing local inflammation and joint damage in murine models of collagen-induced arthritis without causing systemic depletion of beneficial immune cell populations.
