This is a combination therapy of three agents—**napabucasin**, **oxaliplatin**, and **capecitabine**—used in the treatment of advanced solid tumors, particularly gastrointestinal cancers such as colorectal and pancreatic cancer, in clinical trials. Napabucasin is a small molecule cancer stemness inhibitor that primarily inhibits STAT3-driven gene transcription, blocking cancer stem cell proliferation and survival. Oxaliplatin is a platinum-based chemotherapeutic that causes inter- and intra-strand DNA crosslinks, leading to apoptosis in rapidly dividing cells. Capecitabine is an oral prodrug converted to fluorouracil (5-FU), which inhibits DNA synthesis by interfering with thymidylate synthase and also incorporates into RNA. The combination aims to capitalize on the complementary mechanisms: targeting both bulk tumor and cancer stem cell subpopulations, as well as maximizing cytotoxic effects in resistant or recurrent cases[1][2][6].
