Napabucasin + panitumumab is an investigational combination therapy evaluated primarily in patients with metastatic colorectal cancer (mCRC) who have wild-type KRAS. Napabucasin is a first-in-class small molecule inhibitor of cancer stemness, acting by suppressing STAT3-driven gene transcription and targeting pathways implicated in the viability and self-renewal of cancer stem cells. Panitumumab is a fully human monoclonal antibody that targets the epidermal growth factor receptor (EGFR), inhibiting downstream signaling involved in tumor cell proliferation and survival. The combination has been studied for its potential to enhance anti-tumor activity, particularly in patients previously treated with anti-EGFR agents, suggesting napabucasin may sensitize tumors to repeat EGFR blockade. Clinical studies have shown that this regimen can be safely administered together at full doses, with encouraging disease control rates observed in heavily pretreated mCRC populations[4][5][6].
