Overview

Naphazoline is a rapid-acting imidazoline sympathomimetic vasoconstrictor used primarily for the temporary relief of redness, itching, and minor irritation of the eyes and for nasal congestion. It acts by causing the release of norepinephrine at sympathetic nerve endings, which then binds to alpha adrenergic receptors (primarily alpha-1A and alpha-2A adrenergic receptors) on blood vessels in ocular or nasal tissues, resulting in vasoconstriction. This reduces swelling and congestion. Naphazoline is also a mild beta adrenergic receptor agonist, which can lead to rebound vasodilation after its primary effect subsides. It was first developed in 1942 as a nasal decongestant and is now widely available over-the-counter in eye drops and nasal preparations[1][4][5].
