NaPi2b single-target ADCs are a class of antibody-drug conjugates designed to selectively target the sodium-dependent phosphate transport protein 2B (NaPi2b), a multi-pass transmembrane protein encoded by the SLC34A2 gene. NaPi2b is highly expressed in several solid tumors, particularly ovarian cancer and non-small cell lung cancer (NSCLC) adenocarcinoma, while its expression in normal tissues is largely restricted to the lungs, making it a viable target for precision oncology. These ADCs typically consist of a humanized monoclonal antibody conjugated to a potent cytotoxic payload, such as monomethyl auristatin E (MMAE), via a linker system. Upon binding to the target on the tumor cell surface, the ADC is internalized through endocytosis, followed by lysosomal degradation and the release of the payload, which induces cell cycle arrest and apoptosis. While several candidates, such as upifitamab rilsodotin (UpRi), have advanced into late-stage clinical trials, the class has encountered challenges related to the therapeutic window and heterogeneous target expression, leading to the exploration of dual-targeting strategies.
