Napitane (ABT-200) is an investigational small-molecule antidepressant developed by Abbott that functions primarily as a norepinephrine reuptake inhibitor and antagonist of presynaptic noradrenergic alpha-2 receptors, thereby enhancing synaptic noradrenergic transmission.[1][3][5][9][13] Preclinical studies showed that it blocks nicotine-stimulated catecholamine release by inhibiting cation flux through multiple ion channels, and clinical pilot trials evaluated its efficacy versus placebo and versus fluoxetine in major depressive disorder, although it was never commercialized.[1][3][5][9] A scalable, stereospecific manufacturing process starting from 5,6-methylenedioxy-1-tetralone was developed to enable kilogram-scale production for development.[7]
