Naproxen-n-butyl nitrate (HCT-1026) is a nitric oxide-releasing derivative of the non-steroidal anti-inflammatory drug (NSAID) naproxen, belonging to the Cyclooxygenase-Inhibiting Nitric Oxide Donator (CINOD) class. It was developed to provide the anti-inflammatory and analgesic benefits of naproxen while mitigating gastrointestinal and renal side effects through the protective actions of nitric oxide (NO). Beyond its use in inflammatory conditions like osteoarthritis, it was investigated for potential anti-cancer properties, specifically in prostate cancer, where it was hypothesized to inhibit tumor growth through COX inhibition and NO-mediated signaling. Stanford University conducted a Phase II clinical trial (NCT00573443) evaluating naproxen-n-butyl nitrate in combination with high-dose calcitriol for recurrent prostate cancer. However, the study was terminated early due to extreme toxicity, including severe hypercalcemia and renal impairment, which were attributed to the combination regimen.
