Naratuximab is a humanized monoclonal antibody designed to specifically target the CD37 protein, a surface marker found on mature B cells and various B-cell malignancies. It serves as the antibody component of the antibody-drug conjugate (ADC) naratuximab emtansine (also known as Debio 1562 or IMGN529). In this ADC, naratuximab binds to CD37 on cancer cells, facilitating the internalization and targeted delivery of a cytotoxic payload, DM1, to induce cell death. The antibody component itself also contributes to therapeutic effects by inducing antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), thereby showing pro-apoptotic activity. This targeted approach aims to enhance treatment efficacy while minimizing off-target effects.
