This combination therapy consists of narlumosbartmab, a humanized monoclonal antibody targeting the Receptor Activator of Nuclear Factor Kappa-B Ligand (RANKL), and a standard neoadjuvant chemotherapy regimen known as the PKUPH-OS protocol (comprising doxorubicin, cisplatin, methotrexate, and ifosfamide). Developed and investigated by Peking University People's Hospital, this regimen is specifically designed for the treatment of bone-derived malignancies characterized by osteolytic lesions and multinucleated giant cells. Narlumosbartmab works by inhibiting osteoclast activity, which is intended to clarify tumor boundaries and reduce surgical difficulty, while the chemotherapy components provide systemic cytotoxic effects by inducing DNA damage and inhibiting metabolic pathways essential for tumor cell proliferation.
