Overview

Narmafotinib is a highly potent and selective, orally bioavailable small molecule inhibitor of focal adhesion kinase (FAK), a non-receptor tyrosine kinase overexpressed in several cancers, including pancreatic cancer. FAK plays a key role in tumor growth by promoting cell survival, proliferation, migration, chemoresistance, and contributing to fibrosis and immunosuppression within the tumor microenvironment. Narmafotinib has demonstrated promising preclinical activity in models of cancer and fibrosis. It is being developed primarily for advanced pancreatic cancer (currently in Phase 2 trials) but also shows potential for fibrotic diseases such as idiopathic pulmonary fibrosis (IPF), where it has received Orphan Drug Designation from the FDA. The drug is being developed by Amplia Therapeutics.
