Drug intelligence / Profile preview
narmafotinib + gemcitabine + Abraxane is an investigational combination regimen being evaluated as a first-line therapy for advanced or metastatic pancreatic ductal adenocarcinoma (PDAC). The regimen pairs narmafotinib (AMP945), an orally bioavailable, highly selective small-molecule inhibitor of Focal Adhesion Kinase (FAK) developed by Amplia Therapeutics, with the standard-of-care chemotherapies gemcitabine (a nucleoside metabolic inhibitor) and Abraxane (nab-paclitaxel, an albumin-bound nanoparticle formulation of paclitaxel). FAK is overexpressed in pancreatic cancer and contributes to tumor fibrosis, immunosuppression, and chemoresistance. By inhibiting FAK, narmafotinib acts as a priming agent to reduce stromal fibrosis and enhance the penetration and efficacy of the co-administered chemotherapies. The combination is currently being evaluated in the Phase 1b/2a ACCENT clinical trial in Australia and South Korea, where it has demonstrated promising efficacy, including unprecedented complete response rates.
