Drug intelligence / Profile preview
narmafotinib + leucovorin + fluorouracil + irinotecan + oxaliplatin is an investigational combination therapy that was being developed by Amplia Therapeutics for the treatment of advanced or metastatic pancreatic ductal adenocarcinoma. The regimen combines narmafotinib (formerly AMP945), an oral, highly selective small-molecule inhibitor of focal adhesion kinase (FAK), with modified FOLFIRINOX (mFOLFIRINOX), a standard-of-care cytotoxic chemotherapy regimen consisting of leucovorin, fluorouracil, irinotecan, and oxaliplatin. FAK is frequently overexpressed in pancreatic cancer, where it promotes tumor fibrosis, chemotherapy resistance, and immunosuppression. By inhibiting FAK, narmafotinib acts as a stromal priming agent, reducing extracellular matrix stiffness and density to enhance the penetration and efficacy of the co-administered cytotoxic chemotherapies. The combination was evaluated in the Phase 1b/2a AMPLICITY trial (NCT07026279) in Australia and the US; however, in April 2026, Amplia Therapeutics strategically halted recruitment in the AMPLICITY trial due to dose-limiting toxicities associated with the FOLFIRINOX chemotherapy backbone, with no safety concerns identified for narmafotinib itself.
