Narnatumab is a human monoclonal antibody developed as an investigational anticancer agent. It specifically targets the macrophage-stimulating protein receptor (RON; also known as recepteur d'origine nantais), which is expressed on macrophages, epithelial cells, and various tumor types. By binding to RON, narnatumab blocks its interaction with its ligand—macrophage-stimulating protein (MSP)—thereby inhibiting downstream signaling pathways involved in tumor growth and progression. Narnatumab has been evaluated in phase 1 clinical trials for advanced solid tumors but demonstrated limited antitumor activity at tested doses due to suboptimal drug exposure and the prevalence of RON splice variants that are not accessible by large-molecule antibodies[1][4][6][8].
