Overview

Nascertib (ABBV-105) is an orally bioavailable small molecule inhibitor of receptor-interacting serine/threonine-protein kinase 1 (RIPK1), developed by AbbVie. RIPK1 is a critical mediator in signaling pathways that regulate inflammation and programmed cell death, specifically necroptosis and apoptosis, often triggered by TNF-alpha. By inhibiting RIPK1, nascertib is designed to modulate the production of pro-inflammatory cytokines and prevent tissue damage associated with necroptotic cell death. The drug has been primarily investigated for the treatment of autoimmune and inflammatory disorders, most notably rheumatoid arthritis, where it was evaluated in Phase 2 clinical trials as both a monotherapy and in combination with other agents. While it showed potential in early clinical development, recent pipeline updates from AbbVie suggest the program may have been deprioritized or discontinued.
